CHANGES IN CEREBRAL WHITE MATTER, VASCULAR RISK ANDCOGNITION ACROSS THE ADULT LIFESPANbyANDREW R. BENDERMay 2014Advisor: Dr. Naftali RazMajor: Psychology (Behavioral and Cognitive Neuroscience)Degree: Doctor of PhilosophyNumerous studies over the past decade have used diffusion tensor imaging (DTI) toexamine the associations between age, diffusion and anisotropy measures of cerebral whitematter (WM), and cognitive performance. However, few have examined relationshipsbetween intra-individual change in DTI measures of WM and cognitive function. It ispossible that the extant cross-sectional findings are a poor representation of age-relatedchange in WM and cognition. The present study used latent difference-score modeling (LDM)to assess change over two years in DTI indices fractional anisotropy (FA), radial diffusivity (DR), axial diffusivity (DA) and mean diffusivity (MD). In addition, we examined the effectsof WM change on concomitant change in age-sensitive cognitive domains, while controllingfor individual differences in duration of hypertension treatment, a common marker ofvascular risk. A sample of 96 healthy participants spanning the adult lifespan underwent DTIscanning and cognitive testing at baseline and following a two-year delay. Univariate latentdifference score model (LDM) analyses showed cross-sectional associations between DTImeasures and age did not accurately describe actual change in WM. Regions including the

The Relationship between Cortical Blood Flow and Sub-Cortical White-Matter Health across the Adult Age Span

Degeneration of cerebral white matter is commonly observed in aging, and the associated degradation in neural connectivity contributes to cognitive decline in older adults. Vascular dysfunction has been implicated as a potential mechanism for general age-related neural tissue deterioration; however, no prior study has examined the direct relationship between cortical vascular health and subcort...

Longitudinal Changes in White Matter Tract Integrity across the Adult Lifespan and Its Relation to Cortical Thinning

A causal link between decreases in white matter (WM) integrity and cortical degeneration is assumed, but there is scarce knowledge on the relationship between these changes across the adult human lifespan. We investigated changes in thickness throughout the cortical mantle and WM tract integrity derived from T1 and diffusion weighted magnetic resonance imaging (MRI) scans in 201 healthy adults ...

Vascular Contributions to Cognitive Impairment and Dementia

Cerebrovascular disease and Alzheimer disease (AD) lesions are common in older people and accumulate with age. Cerebrovascular lesions can directly reduce cognitive status: vascular dementia is the second most common cause of clinical dementia after AD. In addition, cerebrovascular lesions worsen the impact of AD and other dementia pathologies and may contribute to AD pathogenesis. This spectru...

Untangling vascular cognitive impairment.

See related articles, pages 783–830. Vascular pathology is often the cause of cognitive impairment and dementia. The entities implicated include large cortical infarcts; single strategically-placed or multiple small infarcts, cerebral hemorrhage, cortical changes due to hypoperfusion (eg, hippocampal sclerosis, ischemic-anoxic damage, cortical laminar sclerosis), and a variety of vasculopathies...

Risk factors for vascular dementia: Hypotension as a key point

Physiologically, the cerebral autoregulation system allows maintenance of constant cerebral blood flow over a wide range of blood pressure. In old people, there is a progressive reshape of cerebral autoregulation from a sigmoid curve to a straight line. This implies that any abrupt change in blood pressure will result in a rapid and significant change in cerebral blood flow. Hypertension has of...
